چکیده

Individuals with sarcoidosis were evaluated for preferential usage of T cells with the yb-positive (+) type of T cell antigen receptor. Compared with normal subjects (n = 19), the group with sarcoidosis had increased numbers of CD3+ a(3-negative (-) T cells in the blood (normal, 58±12 cells/ls; sarcoid, 192±45 cells/,Ml, P < 0.05) and in the epithelial lining fluid of the lung (normal, 78±14 cells/Ml; sarcoid, 240±60 cells/Ml, P < 0.04) and a concomitant elevated number of blood and lung CD3+ y5+ T cells, owing to a striking increase in the number of CD3+ -y5+ T cells in a subgroup (7 of 20) of sarcoid individuals. The elevated numbers of sarcoid blood y6+ T lymphocytes were mostly Ti'yA+ and 5TCS1-, a pattern also seen in normal individuals, consistent with the majority of yb+ T cells expressing one '-chain variable region, Vy9. The observation of an increase in the total yb+ T cell numbers in a sarcoid subgroup suggests that various specific stimuli may trigger the expansion of different T cell subpopulations within different groups of individuals with sarcoidosis. Mechanisms of hypergammaglobulinemia in pulmonary sarcoidosis. Site of increased antibody production and role of T lymphocytes.

Pulmonary sarcoidosis is a disorder in which local granuloma formation is perpetuated by activated lung T lymphocytes. The present study suggests that lung T lymphocytes may also play a critical role in modulating local production of antibodies in this disorder. In untreated patients with pulmonary sarcoidosis, the numbers of IgG- and IgM-secreting cells per 10(3) lung lymphocytes are markedly ...

Gamma/delta T lymphocytes in the blood of patients with sarcoidosis.

BACKGROUND Gamma/delta T lymphocytes are thought to have a role in granulomatous immune responses at peripheral sites of antigen contact such as the gut, skin and lung. The aim of this study was to determine if gamma/delta T lymphocytes are increased in the peripheral blood of patients with active sarcoidosis. METHODS Peripheral blood from 21 untreated patients with a new presentation of sarc...

State of the Art Role of CD4 T Cells in Sarcoidosis

Activated pulmonary CD4 T lymphocytes of the Th-1 type are essential for the inflammatory process in sarcoidosis, and IFNproduction is crucial for the characteristic granuloma formation. Both the T cells and their inflammatory mediators may constitute possible targets for immunotherapy. A particular T-cell subset, the T-cell receptor (TCR) AV2S3 bronchoalveolar lavage (BAL) CD4 T cells, is foun...

Enhanced alveolar macrophage-mediated antigen-induced T-lymphocyte proliferation in sarcoidosis.

Expansion of T-lymphocyte numbers is a characteristic feature of the alveolitis of pulmonary sarcoidosis. One mechanism that may influence the numbers of T-lymphocytes in the lung is the process of antigen presentation in which alveolar macrophages, in the presence of antigen, induce T-lymphocytes to replicate. To evaluate this process in sarcoidosis, alveolar macrophages were obtained by bronc...

Phenotypic analysis of lymphocytes and monocytes/macrophages in peripheral blood and bronchoalveolar lavage fluid from patients with pulmonary sarcoidosis.

BACKGROUND The granulomatous inflammation in sarcoidosis is driven by the interplay between T cells and macrophages. To gain a better understanding of this process the expression by these cells of cell surface activation markers, co-stimulatory molecules, and adhesion molecules was analysed. METHODS CD4+ and CD8+ T lymphocytes from peripheral blood (PBL) or bronchoalveolar lavage (BAL) fluid,...
